摘 要:在辅酶NADH存在下,以硝基还原酶催化还原对-硝基苯酚(4-nitrophenol,4-NP)和2,4-二硝基苯酚(2,4-dinitrophenol,DNP)。通过紫外-可见分光光度法、高效液相色谱法和质谱法分析还原反应产物,确定4-NP被还原生成了对-羟氨基苯酚和对-氨基苯酚,DNP被还原生成了H--DNP。计算得出每毫克酶在有氧条件和无氧条件下对硝基苯酚的还原率分别为4.176和4.172 nmol/h,可见有氧条件和无氧条件下具有相似的还原率,所以实验所用硝基还原酶属于对氧不敏感型(TypeⅠ)。由上述结论推测出硝基苯酚的酶催化还原机制。The reductions of 4-nitrophenol (4- NP) and 2,4-dinitrophenol (DNP) catalyzed by nitroreductase in the presence of NADH are investigated. 4-Aminophenol and 4-hydroxylaminophenol are found in the reductive products of 4 - NP. H^- - DNP is in the reductive products of DNP. The similar reducing ratios of nitro phenols are obtained both under aerobic and anaerobic conditions, and the results indicate that an oxygen-insensitive reaction is proceeded in the reductions of nitro phenols. This enzyme can be classified as an oxygen-insensitive nitroreductase. Moreover, the reductive products of nitro phenols are determined by UV - vis, HPLC and MS.
