摘 要:目的:探讨益气活血方和补肾生髓方对局灶性脑缺血再灌注损伤大鼠恢复期缺血半暗带脑源性神经营养因子(BDNF)和碱性成纤维细胞生长因子(bFGF)表达的影响。方法:采用线栓法阻塞大鼠左侧大脑中动脉,复制局灶性脑缺血再灌注模型,缺血2h分别再灌注1w、2w、3w,免疫组化SABC法分别检测缺血半暗带BDNF和bFGF的表达。结果:假手术组BDNF和bFGF弱阳性表达;再灌注各时间点,模型组BDNF和bFGF表达的平均吸收光密(OD值)升高,与假手术组比较有显著性差异(P<0.01,在3w时bFGF除外);在多个时间点益气活血方组、补肾生髓方组BDNF和bFGF表达的OD值均较模型组升高;两中药复方组间比较,在1w、2w,BDNF表达的OD值益气活血方组高于补肾生髓方组;第3w,补肾生髓方组高于益气活血方组,各时间点bFGF表达的OD值,两中药复方组间无显著差异。结论:益气活血方、补肾生髓方能通过促进缺血半暗带BDNF和bFGF的表达,抗脑缺血后损伤,有利于脑缺血后神经功能的恢复。Objective: To investigate the effect of Yiqihuoxue prescription and Bushenshengsui prescription on BDNF and bFGF expression in convalescent period Rats' Cerebral Ischemia-reperfusion penumbra. Methods: The focal ischemic model of middle cerebral artery occlusion (MCAO) in rats were made by inserting nylon surgical thread into the left middle cerebral artery. The middle cerebral arteries(MCA) of rats were occluded for 2 hours and reperfused for 1,2,3 weeks respectively. The expression of BDNF and bFGF in convalescent period was detected by SABC immunohistochemistry method. Result: In the normal brain tissues, only a few BDNF and bFGF immunofluorescence staining positive cells were found. In the model group, the BDNF and bFGF in convalescent period expression significantly increased at 1w,2 w,3w compared with those in sham operation group(P〈0.01, except the bFGF at 3w).In the Yiqihuoxue and Bushenshengstti prescription treatedgroups, the BDNF and bFGF in convalescent period expression increased at 1w,2w ,3w compared with those in model group; At 1w,2w the BDNF expression in the Yiqihuoxue prescription treatedgroup increased compared with those in Bushenshengsui prescription. But at 3w the BDNF expression in the Bushenshengsui prescription treatedgroup increased compared with those in Yiqihuoxue prescription. At 1w,2w ,3w the bFGF expression in the Yiqihuoxue and Bushenshengsui prescription treatedgroups had no notable difference. Conclusion: The Yiqihuoxue prescription and Bushenshengsui prescription contribute to focal cerebral ischemia-reperfusion rats'synaptic remodeling and axonal regeneration by promoting the expression of BDNF and bFGF in convalescent period, and benefit nerve functional recovery after cerebral apoplexy.
