摘  要:[目的]探讨低能离子注入对烟草反转录转座子Tnt1多态性的影响。[方法]利用TRAP技术检测离子束辐照后的烟草的反转录转座子Tnt1的多态性。[结果]随着离子束辐照剂量的增大,发芽率基本成线性减少的趋势,但在注入过程中的真空状态对种子的发芽率基本没有影响。经过离子束辐照后,烟草植株的生理特征展示出一些较明显的变化。烟草的反转录转座子Tnt1发生了转座。通过TRAP分析,Tnt1展示出一定的多态性。利用TRAP技术,通过对样本DNA的扩增,对出现的特异性条带进行测序,经BLAST搜索,没有发现与Tnt1匹配率较高的序列,初步判断其为功能未知的序列。[结论]该研究为揭示离子束的诱变机理提供了依据。[ Objective ] The aim was to discuss the influence of low energy ion implantation on the polymorphism of tobacco retrotransposon Tnt1. [ Method] The TRAP technology was used to detect the polymorphism of retrotransposon Tnt1 in the tobacco irradiated by ion beam. [ Result ] The germination rate was diseased linearly with the increasing of ion beam irradiation dosage, but during implantation process, the vacuum condition had no influence on the seed germination rate. After irradiated by ion beam, the physiological characteristics of tobacco plants showed some more obvious changes. The tobacco retrotransposon Tntl made transposition. Through TRAP analysis, Tnt1 showed certain polymorphism. The sample DNA was amplified by TRAP technology and the specific band was sequenced. The BLAST showed that there was no higher matching rate sequence with Tnt1. The specific band was judged as unknown functional sequence. [ Conclusion] The research provided a basis for revealing the mutation mechanism of ion beam.
