摘  要:腐植酸是一种深褐色荧光的有机高分子复合物,它是由酚、醛、酸组成。在大鼠腹腔注射腐植酸能诱导大鼠睾丸形态的变化,包括曲细精管的退化,塞尔托利细胞和精原细胞数目的减少和精子细胞的丢失。有人认为塞尔托利细胞可能与睾丸萎缩的过程有关。在本研究中,使用了老鼠的塞尔托利细胞系-TM4研究腐植酸对塞尔托利细胞的影响和腐植酸诱导睾丸萎缩的机制。发现在与腐植酸接触的第1天至第4天TM4细胞生长减慢。使用流式细胞仪分析腐植酸处理的TM4细胞中DNA的含量,发现TM4细胞没有G_1的次高峰,这表明TM4细胞对程序性细胞的死亡没有作用。然而,大部分的TM4细胞在G_1期处于静止状态。在腐植酸处理4天后,处于G_1期的TM4细胞的比例从36%增加到84%。腐植酸处理的TM4细胞的Western免疫印记分析表明:当p27^(kipl)蛋白的表达增加时细胞周期蛋白D_1的表达在减少。研究结果表明,腐植酸诱导睾丸萎缩与对塞尔托利细胞的生长受到抑制有一定的关系。这个模型可能对环境试剂导致的睾丸萎缩的研究有作用。Humic acid(HA) is a fluorescent deep brown organic, polymeric compound composed of hydroxybenzene, aldehyde and organic acid. Intraperitoneal injection of HA in rats could induce morphological change of testicle, including degeneration of seminiferous tubule, reduction of the number of Sertoli cells and spermatogonia, and a loss of spermatids. It was suggested that Sertoli cells may be involved in the progression of testicular atrophy. In this study, a rat Sertoli cell Line, i.e., TM4, was used to investigate the effect of HA on Sertoli cells and the mechanism of testicular atrophy induced by HA. The results showed that the growth rate of TM4 ceils was reduced in 1 to 4 days after exposed to HA. No sub-G1 peak was found though analyzing DNA content in TM4 ceils treated with HA by using FACS method, which indicated that TM4 cells did not commit to the programmed cell death. However, a large proportion of TM4 cells were arrested at the G1 phase, and the percentage of TM4 cells increased from 36% to 84% after treated with HA for 4 days. Western blot assay of TM4 cells treated with HA showed that the expression of cyclin D1 protein decreased, while the expression of p^27kip1 protein increased. These results suggested that HA-induced testicular atrophy was related partly to an inhibitory effect on Sertoli cells growth. This model may be useful to investigate the inducing effect of environmental agents on testicular atrophy.
