摘 要:利用柚木继代组培试管苗,调整组织培养基本培养基(Ms)配方中的Ca^2+浓度和pH值,采用随机区组试验设计,开展了5个Ca^2+浓度(0,3,6,9,12mmol·L-1)和4个pH值水平(pH值4.0,pH值5.0,pH值6.0,pH值7.0)双因素试验,目的是探讨不同Ca2+浓度与pH值对柚木组培苗生长和矿质营养吸收的影响.结果表明:幼苗的高生长及生物量积累与Ca^2+浓度呈正相关,最适Ca^2+浓度为9mmol·L-1;pH值对苗高生长及愈伤组织生物量积累有显著影响,适宜pH值为6.0;茎叶的钙含量随Ca^2+浓度的增加而增加,与培养基的pH值高低无必然关系;在pH值6.0培养条件下,低至中等Ca^2+浓度(3~6mmol·L^-1)有利于幼苗对N,P,Mg,Mn,zn,B,Cu营养元素的吸收,较高的Ca^2+浓度有利于提高钾离子吸收的选择性及提高N吸收量,幼苗对铁离子的吸收随着Ca^2+浓度的增加而下降.A two-factor experiment was undertaken by employing randomized block design to test the growth and nutrient-uptake responses of teak (Tectona grandis) seedlings in vitro to 0, 3, 6, 9 and 12 mmol· L-1 of calcium concentrations and pH treatments of subculture medium. Resuhs indicate that the height increments and dry weights of leaf and stem are positively related to the Ca^2+ concentration, the optimal of which is 9 mmol· L-1; that the height growth is significantly affected by pH values, the optimal of which is 6 ; that the total Ca content in leaf and stem is enhanced with the increase of Ca^2+eoncentration in subculture medium, having no necessary relation with low pH values; that 3-6 mmol · L-lof Ca2+eoncentration is favorable for teak tube seedlings to uptake N, P, Mg, Mn, Zn, B and Cu elements and a high Ca^2+concentration, 9 mmol ·L-1 , is beneficial for K-ion uptake; and that Fe-uptake is markedly restrained with the increase of Ca^2+ concentration.
