摘　　要：目的探讨蛋白酶体α6亚单位(proteasome subunit alpha type 6 gene,PSMA6)基因多态性与甘肃东乡族人群糖尿病及体质量指数(BMI)和血压的相关性。方法采用聚合酶链反应对117例甘肃东乡族2型糖尿病患者(T2DM组)和144例同期正常糖耐量者(NGT组)PSMA6基因-8C→G酶切位点进行多态性分析;同时,用标准的方法测量所有研究对象的身高、体质量和血压等相关指标。结果PSMA6基因-8C→G位点多态性与甘肃东乡族人群2型糖尿病有关(P=0.016)。BMI≥25组与BMI<25组基因型及等位基因频率分布差异有统计学意义(P=0.001)。比较T2DM组与NGT组收缩压、舒张压和腰臀比(WHR)差异亦均有统计学意义(P<0.01)。比较两组基因型之间的差异时可以发现,NGT组的收缩压和舒张压各基因型之间差异有统计学意义(P<0.05),而T2DM组中只有舒张压各基因型之间差异有统计学意义(P<0.05)。结论PSMA6基因-8C→G位点多态性与甘肃东乡族人群糖尿病、血压及BMI有关联。Objective To study the association of the proteasome subunit alpha type 6 gene（PSMA6）-8C→G polymorphism with body mass index（BMI） and blood pressure in diabetic patients of Dongxiang people. Methods The genotypes of -8C→G variant in PSMA6 gene were determined by polymerase chain reaction-restriction fragment length polymorphisms assay in 117 patients with type 2 diabetes mellitus（T2DM group） and 144 individuals with normal glucose tolerance（NGT group）. And meanwhile, body height, body mass, blood pressure and others were measured by standard technique. Results The genotype or allele frequency of T2DM group was significantly different from that of NGT group （ P = 0. 016）, and between BMI~ 25 and BMI〈 25 subjects （ P = 0. 001）. There were significant differences between T2DM group and NGT group about the levels of systolic blood pressure（SBP）, diastolic blood pressure（DBP） and waist/hip ratio（WHR）, respectively（ P 〈0.01）. However,in the subgroup analysis, the DBP level of the CG＋-GG genotype was significantly higher than that of the CC, respectively, in both T2DM group and NGT group（ P 〈0.05）. The SBP was only higher in NGT group （P 〈0.05）. Conclusion The PSMA6--8C→G polyrnorphism is associated with T2DM,blood pressure and BMI in Dongxiang people.
