摘  要:目的探讨幽门螺杆菌(Helicobacter pylori,Hp)对感染细胞增殖凋亡及细胞周期蛋白抑制因子p27^kip1表达的影响。方法不同浓度脚国际标准株NCTC11637分别感染胃上皮细胞株SGC-7901,测定p27^kip1蛋白及mRNA的表达、细胞增殖指数和凋亡率。结果(1)Hp在3.4×10^4~1.9×10^5CFU/ml浓度时,SGC-7901的增殖指数随却浓度增高而显著增高,而在Hp≥4.8×10^6CFU/ml时明显降低;(2)Hp≥9.6×10^5CFU/ml时,细胞凋亡率呈浓度依赖性显著增高(P〈0.01);(3)细胞p27^kip1 mRNA表达阳性率无显著变化,而p27^kip1蛋白表达阳性率呈脚浓度依赖性显著降低(P〈0.05),与细胞凋亡率呈负相关。结论却感染可直接导致胃上皮细胞增殖与凋亡的紊乱,影响细胞周期调控蛋白p27^kip1的表达。Objective To explore the influence of Helicobacter pylori (Hp) on the proliferation, apoptosis and expression of p27^kip1 protein of gastric epithelial cells in vitro. Methods SGC-7901 cell proliferation was examined by flow cytometry after incubation with different concentration of NCTC11637. The effect of NCTC11637 on cell apoptosis was also evaluated by flow cytometry. And the expression of p27^kip1 of gastric epithelial cells was detected by immunohistochemical analysis and in situ hybridization, respectively. Results Cell proliferation was enhanced when co-incubated with the Hp in low concentration (3.4 ×10^4 to 1.9 ×10^5 CFU/ml) but inhibited in higher concentration (≥4.8 ×10^6 CFU/ml). However, cell apoptosis was increase when co-incubated with the Hp in high concentration ( ≥9.6 ×10^5 CFU/ml) showing concentration dependent picture. In addition, the co-incubation of SGC-7901 with Hp led to decrease of the expression of p27^kip1 protein but not mRNA in a Hp concentration dependent way. Conclusion Hp could effect the gastric epithelial cells apoptosis and proliferation directly and influence the expression of p27^kip1 protein which might facilitate gastric carcinoma.
