摘  要:目的:研究藻酸钙水凝胶-兔脂肪干细胞(ADSCs)复合物体内异位成骨能力,探讨其作为可注射组织工程骨的可能性。方法:使用藻酸盐溶液以2×107/ml的终细胞密度悬浮ADSCs,形成凝胶后注射于裸鼠背部皮下,8周后取材,通过大体标本观察、CT扫描、组织学检查,观察骨组织的形成情况。结果:皮下形成质地较硬的结节,阻射密度较高,组织学及电镜检查证实有骨样结构形成,单纯藻酸钙凝胶注射无新骨形成。结论:ADSCs与藻酸钙水凝胶可以分别作为种子细胞和支架载体构建可注射的骨组织。Objective To study the osteogenesis ability of rabbit adipose-derived stem cells (ADSCs) and the probability that they may be used as seed cells for bone tissue engineering. Methods The ADSCs- hydrogel complexes were injected subcutaneously into the backs of the nude mice. CT scans, gross, histological, and SEM examinations were performed to evaluate the harvested specimens. Results Aight weeks after injection hard nodus formed subcutaneously. Three-dimensional CT images exhibited evident ectopic mineralization. The formation of new bone was also confirmed by histological and SEM examination. Conclusions New bone could be regenerated through the injection of alginate hydrogel seeded with ADSCs, this method might have great potential of clinical use.
