摘  要:目的观察自身免疫性肝炎(autoimmune hepatitis,AIH)患者和健康供肝标本中人高迁移率族蛋白B1(high mobility groupbox-1 protein,HMGB1)的表达情况,探讨HMGB1蛋白与AIH发病的相互作用。方法免疫组织化学方法观察55例AIH患者肝穿组织标本和10例健康肝移植供肝标本中HMGB1蛋白表达情况。结果AIH患者的肝穿标本与健康肝脏标本相比,肝细胞中HMGB1阳性表达明显增多,HMGB1阳性颗粒主要分布在肝细胞核和Kuffer细胞核中,在点灶状坏死区和界面炎症区浸润的炎性细胞细胞核和细胞浆内可见HMGB1阳性颗粒。结论HMGB1是AIH发生发展中一个重要的炎症蛋白。Objective To investigate the potential role of human high mobility group box-1 protein (HMGB1) in pathogenesis of autoimmune hepatitis (AIH). Methods Specimens from 55 AIH liver biopsies and 10 normal livers were stained and the expression of HMGB1 was observed by immunohistochemistry method. Results Markedly correlation was proved that HMGB1 expressed much more strongly in AIH hepatocytes than that in healthy control. Data showed that HMGB1 protein was mainly found in nucleus of hepatocyte and Kuffer cells, as well as in the inflammatory cells which aggregated in inflammatory and necrotic area. Conclusion HMGB1 is an important inflammatory mediator in pathogenesis of AIH, which may shed light on understanding of AIH and provide a potential target for AIH control.
