摘 要:目的探讨阿魏酸钠(SF)对高糖诱导的原代培养系膜细胞(GMC)增殖的作用及其机制。方法原代培养肾小球系膜细胞,分为正常组、甘露醇组、高糖组、高糖加不同浓度SF组,分别于处理后24、48h收集细胞。用MTT法检测细胞增殖;流式细胞术测定cyclinD1和细胞周期;免疫细胞化学检测p53蛋白的表达。结果48h内,高糖促进GMC增殖,使GMC由G1期进入S期细胞比例增高;同时cyclinD1蛋白表达上调而p53蛋白的表达减少;SF能明显对抗高糖的这种作用,其作用随SF浓度的增加、作用时间的延长而加强。结论SF可能通过抑制cyclinD1蛋白表达和促进p53蛋白生成,从而抑制高糖诱导的GMC增殖。Objective To investigate the effects and mechanism of sodium ferulate (SF)on the proliferation of mesangial cells induced by high glucose. Methods The primary glomerular mesangial cells were divided into normal, mannitol, high gloucose and high gloucose plus different concentration SF groups. Cells were collected at 24 and 48 h. Cell proliferation was measured by MTT assay, and cell cycle and cyclinD1 were measured through flow-cytometry, p53 protein expression was detected by immunecytochemistry. Results High glucose significantly promoted mesangial cell proliferation and up-regulated the expression of cyclinD1, and down-regulated expression of p53 within 48 h. SF inhibited mesangial cell proliferation induced by high glucose with concentration and time-depended manner. Conclusions SF can inhibit proliferation of mesangial cells induced by high glucose by suppressing the cydinD1 expression and increasing the level of p53 protein possibly.
