羌银解热汤治疗流行性感冒的基础研究

摘　　要：目的:研究羌银解热汤对小鼠感染甲1型流感病毒FM1株的抑制作用和对小鼠的保护作用;观察羌银解热汤对感染H3N2型人流感病毒的狗肾传代细胞(Madin-Darby canine kidney,MDCK)的保护作用以及该汤药对H3N2型人流感病毒的直接灭活作用。方法:利用甲1型流感病毒FM1株感染小鼠为动物模型,观察羌银解热汤在小鼠体内的抗流感病毒作用,并与银翘散汤剂、荆防败毒散汤剂作以对比。用H3N2型人流感病毒感染体外培养成单层生长的MDCK细胞,观察羌银解热汤体外抗流感病毒的作用。结果:羌银解热汤可以有效抑制病毒引起的小鼠肺部病变、降低肺指数(P<0.05),改善小鼠组织病理变化,提高对感染流感病毒小鼠的死亡保护率(P<0.05),降低流感病毒诱导的血清干扰素IFN-α水平,其效果优于银翘散汤剂、荆防败毒散汤剂。体外实验显示羌银解热汤对H3N2型人流感病毒并无直接灭活作用,对MDCK细胞的保护作用并不明显。结论:实验结果表明羌银解热汤具有良好的体内抗流感病毒感染作用,其作用机理不能用该汤药的直接灭活流感病毒来解释。Objective：To observe the protective effects of Qiang Yin JieRe Tang on the mice infected with influenza A virus FM1 strain and MDCK cells infected with human influenza virus H3N2. Methods ：120 mice were randomly divided into eight groups with 15 mice in each group, normal control group, model group, group with ribavirin, group with YinQiao San, group with JingFang BaiDu San, group with QiangYin JieReTang. Normal control group excluded, all mice in the other groups were infected with influenza A virus FM1 strain. The impacts on lung index and death rate of mice were observed in the mice infected with influenza A virus. Madin-Darby canine kidney（MDCK） cells infected with human influenza viruses were randomly divided into three groups ,group normal control, group with ribavirin, group with QiangYin JieRe Tang, the protective effects of QiangYin JieRe Tang on MDCK cells were observed. Results ： QiangYin JieRe Tang at the concentrations of 4g/100g and 2g/100g for the treatment markedly inhibited the lung consolidation of mice pneumonia caused by the infection of influenza virus, （ P 〈 0. 05 ）. Compared with the model group, the death rate of mice of QiangYin JieRe Tang at the, concentrations of 4mL/100g was lower, （ P 〈 0. 05）. The protective effects of QiangYin JieRe Tang on MDCK cells infected with human influenza rims H3 N2 was not observed. Conclusion：QiangYin JieRe Tang can markedly inhibiting influenza A virus FM1 strain in the mice infected with influenza virus.
