博落回植株地上部分各器官生物碱含量的研究

摘　　要：目的:探讨博落回植株地上部分各器官生物碱的含量,为进一步开发利用博落回植物资源提供科学依据。方法:应用正交设计的方法,以浸膏得率和生物碱含量为考察指标,对博落回生物碱的提取条件进行优选。然后,在优选条件下对同一生长环境中同一时期采集的博落回植株茎、叶、果实各部位进行生物碱提取及其含量测定。结果:各考察因素对博落回生物碱提取工艺的影响程度是溶剂量倍>药材预浸时间>提取次数/提取时间>提取温度。博落回生物碱提取的最佳条件为A1B1C1D1。在相同的实验条件下,同一生长时期、同一生长环境中的博落回植株地上部分各器官生物碱的含量是果实最高,叶次之,茎最少。结论:在pH值2的85%乙醇溶液中,博落回生物碱提取的最佳工艺条件是用5倍量固液比将药材在溶剂中预浸24h,提取2次,每次20min,提取温度为20℃。博落回植株地上部分各器官中生物碱的含量是不同的。Objective： To study the total alkaloids content on different parts of Macleaya cordata plant to provide scientific basis for further expeitation and utilization on its resourses. Methods： By adopting the way of orthognnal design and taking the extract ratio and alkaloids content as an observative indication, we select the extraction methods by optimum seeking. On this condition, we recover alkaloids and determine its contents from the stem ,leaf and fruit of Macleaya eordata plant in the same growing situation and the same period. Results： Each observative factor will affect the technological process for its extraction in the proportion like solvent volume 〉 preliminary dipping duration 〉 extraction duration/extraction times 〉 extraction tempreture. The best condition for the extraction of alkaloids is A1 B1 C1 D1. On the same experimental condition,collected in the same growing situation and the same period the total alkaloids content in the fruit is verified to be the richest, the leaf, second and the stem, the lowest. Conclusion：The best technological process for alkaloids extraction is to soak into 85% of pH value 2 of ethanol concentration for 24 hours with 5-fold solid-liquid ratio, extracting for 2 times and 20 minutes for each time with the tempreture at 20℃. The alkaloids content on every part of Macleaya Cordata plant is of difference.
