寿胎丸对自然流产模型小鼠母胎界面γ-干扰素和白介素-10基因表达的影响

摘　　要：目的:探讨寿胎丸对反复自然流产模型小鼠蜕膜组织Th1/Th2细胞因子γ-干扰素(IFN-γ)和白介素-10(IL-10)mRNA表达的影响。方法:设立正常妊娠与自然流产模型,并将自然流产模型小鼠随机分为4组,分别为模型组,寿胎丸低、中、高剂量组,应用原位杂交法检测孕14天各组蜕膜组织IFN-γmRNA和IL-10mRNA的表达。结果:IFN-γmRNA和IL-10mRNA在各组均有表达,与正常妊娠组相比模型组IFN-γmRNA明显升高(P<0.01),IL-10mRNA表达明显降低(P<0.01);与模型组相比寿胎丸低、中、高剂量组IFN-γmRNA表达明显降低(P<0.01),IL-10mRNA表达明显升高(P<0.01)。结论:寿胎丸可能通过降低自然流产小鼠模型IFN-γmRNA表达、升高IL-10mRNA表达,从而调控Th1/Th2型细胞因子的平衡,以达到治疗反复自然流产的目的。Objective：To study the effect ShouTai Pill makes on the expression of Th1/Th2 cytokines：IFN -γnRNA and IL - 10mRNA at the decidua tissues of mice with recurrent spontaneous abortion. Methods ： Build mice models with normal pregnancy and with spontaneous abortion,divid the mice with spontaneous abortion into 4 groups randomly：Model group,ShouTai Pill low dose group,ShouTai Pill middle dose group,ShouTai Pill high dose group. Detect the expressions of IFN -γmRNA and IL - 10mRNA at the decidua tissues perganting for 14 days of each group by in situ hybridization. Resuhs：IFN-γmRNA and IL-10mRNA all has expressions in each group,compared with the group with normol pregnancy, the expression of IFN -γmRNA obviously increased in model group （ P 〈 0. 01 ）, the expression of IL - 10mRNA obviously decreased （P 〈 0. 01 ） ;compared with the model group, in ShouTai Pill low, middle and high dose group, the expression of IFN -γmRNA obviously decreased （ P 〈 0.01 ）, and the expression of IL - 10mRNA obviously increased（ P 〈 0. 01 ） ,but compared with the group with normol pregnancy, there was no obvious differences （P 〈 0. 01 ）. Conclusion： ShouTai Pill may regulate the blance of Th1/Th2 cytokines to attain the purpose of curing recurrent spontaneous abortion, by decreasing the expression of IFN -γ/mRNA and increasing the expression of IL - 10mRNA on the mice models with spontaneous abortion.
