摘  要:目的:采用二次回归通用旋转试验设计优化土茯苓总黄酮(TFSG)的提取工艺。方法:根据Box-Be-hnken的中心组合试验设计原理,在单因素试验的基础上采用四因素三水平的响应曲面分析法,依据回归分析确定各工艺条件的影响因子,以TFSG得率(%)为响应值作响应面和等高线图。结果:TFSG超声波辅助提取效果的影响因素由大到小顺序为:乙醇浓度>超声功率>超声时间>料液比。优化后的工艺参数为:乙醇浓度57%,超声功率500W,超声时间34min,料液比1:21,响应值达到2.32%。结论:本文所建立的数学模型极其显著(P<0.01),可用来对TFSG的提取工艺进行分析和预测。Objective:To optimize the extraction technique of total flavonoids from Smilax glabra Roxb. (TFSG) by quadratic regression general rotary unitized design. Methods :The method of response surface analysis with 4 factors and 3 levels was adopted according to the Box-Behnken center-united experimental design prineiples and one-factor tests. Then the factors influencing the technological parameters were determined by means of regression analysis. Response surface and contour were finally graphed with the extraction rate of TFSG( % )as the response value. Results :The factors which influence the extraction rate of TFSG aecording to priority was ethanol concentration,ultrasonic power, ultrasonic extraction time, and solid/llquid ratio. The optimum eunditions of TFSG extraction were: the ethanol concentration of 57%, the ultrasonic power of 500 W,the ultrasonic extraction time of 34 rain,the solid/liquid ratio of 1:21 ,and the extraction rate of TFSG was up to 2. 32 %. Conclusion: The mathematical model that the present study established was significant ( 19 〈 0.01 ) ,and it might be applied to analysis and prediction the extraction technique of TFSG.
