摘 要:目的观察高压氧(HBO)对新生大鼠缺氧缺血性脑损伤(hypoxia—ischemia brain damage,HIBD)后脑组织存活素(survivin)及细胞凋亡蛋白酶-3(caspase-3)表达的影响,并探讨其保护机制。方法144只7d龄SD大鼠随机分为对照组(n=16)、新生HIBD组(n=64)及HBO治疗组(n=64)。采用经典的Rice法制作缺氧缺血性脑损伤模型,应用Epics XL流式细胞仪检测细胞凋亡情况,并应用免疫组织化学法检测Survivin和Caspase-3表达变化。结果与对照组相比,HIBD组细胞凋亡率及Survivin、Caspase-3表达水平明显升高,组间比较差异有统计学意义(P〈0.05)。通过相关性分析,HBO治疗可促进存活素表达,其表达水平和caspase-3表达负相关。结论HBO可能通过增加抗凋亡因子Survivin表达来抑制Caspase-3激活,减少缺氧缺血性脑损伤后细胞凋亡,发挥神经保护作用。Objective To explore the effect of hyperbaric oxygen (HBO) on the expression of survivin and caspase-3 following hypoxia-ischemia brain damage (HIBD) in neonatal rats, and the mechanism involved in the protection of the neural system by HBO. Methods One hundred and forty-four ( aged seven days) rats were randomly divided into three groups, i.e. the control group (n = 16), the HIBD group (n = 64) and the HBO group (n = 64). An experimental HIBD model was developed by the Rice method. Following damage, samples were obtained for measuring apoptosis of cells with the Epics XL flow cytometer and survivin expression and easpase-3 proteins were measured with immuno-histochemistry. Results When compared with that of the control group, the number of apoptotie cells, survivin, and caspase-3 immunopositive cells all increased significantly in the HBID group, and statistical differences could be seen between groups ( P 〈 0.05 ). Analyses showed that HBO could significantly enhance the expression of survivin, with negative correlation between the level of survivin expression and the activity of caspase-3. Conclusions Increased expression of survivin by HBO might be one of the mechanisms involved in the regulation of easpase-3 and inhibition of neural cell apoptosis in cortex, which might be protective to neurons.
