摘  要:目的建立同时测定十味活血丸中升麻苷和甘草苷含量的HPLC方法。方法采用HPLC法,色谱柱为Diamonsil C18(200mm×4.6mm,5m),流动相为乙腈-体积分数为0.5%的冰醋酸溶液(体积比为15:85),流速:1.0mL·min^-1,检测波长:276nm,柱温:40℃。结果十味活血丸中升麻苷和甘草苷与其他成分分离良好。升麻苷的质量浓度在2.86—28.6mg·L^-1(r=0.9997)、甘草苷的质量浓度在0.774—7.74mg·L^-1(r=0.9999)内与峰面积呈良好的线性关系.升麻苷和甘草苷的平均回收率分别为99.2%和99.0%,RSD(n=9)分别为1.8%和1.6%。结论该法可用于十味活血丸中升麻苷和甘草苷的含量测定。Objective To establish an HPLC method for simultaneous determination of prim-O-glucosylcimifugin and liquiritin in Shiweihuoxue (traditional Chinese medicine)pill. Methods The separation was performed on a Diamonsil C18(4. 6 mm × 200 mm,5 μm)column. The mobile phase was composed of acetoniWile and 0. 5 % (th) glacial acetic acid solution at the rate of 15:85. The column temperature was set at 40 ℃ and the flow rate was 1.0 mL·min-1i, with UV detection at 276 nm. Results The numbers of theoretical plates calculated for prim-O-glucosylcimifugin and liquiritin were 4 600 and 5 500 ,respectively. The regression equation of prim-O-glucosylcimifugin was A = 8. 910 × 10^3 p + 1. 812 ×10 ( r = 0. 999 7 ), which was linear over the ranger of 2. 86-28.60 mg·L^-1. The regression equation of liquiritin was A = 2. 548 ×10^4 p +8.741 × 10 ( r = 0. 999 9), which was linear over the ranger of 0. 774 - 7.74 mg·L^-1. The average recovery was 99. 2% (RSD = 1.8% )for prim-O-glucosylcimifugin ,99.0% (RSD = 1.6% )for liquiritin. Conclusions The method is simple and accurate, and can be used as the method for quality assessment of Shiweihuoxue pill.
