摘 要:目的:观察灵芝对糖尿病大鼠肾组织氧化应激的影响。方法:30只雄性SD大鼠随机分为3组:正常对照组(C组),糖尿病未治疗组(DM组),糖尿病灵芝(1.2g.kg-1)治疗组(GL组)。腹腔注射链脲佐菌素(STZ)诱导大鼠造成糖尿病模型,GL组用灵芝进行治疗性灌胃。8周后,所有大鼠一次性处死。检测肾组织丙二醛(MDA)的含量、肾组织超氧化物歧化酶(SOD)及过氧化氢酶(CAT)与谷胱甘肽过氧化物酶(GSH-Px)活性;同时进行光镜、透射电镜观察各组大鼠肾皮质病理改变。结果:DM组大鼠出现糖尿病肾病(DN)典型的病理改变。GL组肾组织MDA含量明显低于DM组,SOD、CAT、GSH-Px活性明显高于DM组(P<0.01),病理改变好转。结论:灵芝可能通过抑制肾组织氧化应激,起到对糖尿病大鼠肾脏的保护作用。Objcective: To investigate the effect of Ganederma lucidum (GL) on the oxidative stress in the kidney in diabetic rots. Methods: Thirty Sprague - Damly rats were randomly divided into three groups: normal control group ( C group ), diabetes mellitus group (DM group) and diabetic group treated with GL (1.2g·kg^-1)(GL group) . Diabetic model was induce by streptezocin (60mg·kg^-1 ) and remedial peffusing stomach by administrating GL was done on the GL group. Rats were killed at 8 weeks and kidneys were taken out.The activities of antioxidant enzymes including maleie dialdehyde (MDA), superoxide dismutsse (SOD ), catalase (CAT) and glutathione peroxidase (GSH- Px) in kidney tissure was determined.Renal cortex tissue morphology were obwerved by light microscope and transmission electron microscope.Results: Levels of MDA were higher and SOD, CAT as well as GSH-px activities wree significantly lower in glomeruli in DM group rats than those in C group. GL administration could all remit these changes ( P 〈 0.01) .Meanwhile, the pathologic changes of glomeruli were serious in DM greup.The abnormal expressions of these proteins and pathologic changes were dramafically improved in diabetic rats by GL treatment. Conclusion: GL could exert protective effects on diabetic nephropathy, partly through correlation with the suppression of increase in oxidative stress in diabetic rats.
