马立克氏病毒感染致鸡胚成纤维细胞4号染色体上微卫星不稳定性研究

摘  要:应用微卫星技术对感染马立克氏病毒5 d后的32个鸡胚成纤维细胞(CEF)4号染色体上的20个微卫星标记进行PCR扩增,扩增产物采用变性聚丙烯酰胺凝胶电泳,银染检测。试验结果显示:20个微卫星标记有18个表现出不同程度的微卫星不稳定性,其中ABR0622发生率最高,为37.50%;MCW 0174、ADL0266和ABR0382的发生率较高,均为31.25%。在32组样品中有30个检测到微卫星不稳定性,其中11号样品发生率最高,为45.00%,其次是3号样品为35.00%。结论:CEF感染马立克氏病毒后出现微卫星不稳定性现象。The chicken embryo fibroblast (CEF) were infected with Marek's disease virus (MDV), and five days later , 32 sampies (half of each sample infected with MDV, the other half as control) were detected using 20 microsatellite markers on chicken Chromosome 4 by polymerase chain reaction, whose products were tested by denatured polyacrylamide gel electrophoresis with silver staining. Microsatellite instability (MSI) was found in 18 out of 20 markers, the sensitivity of ABR0622 was 37.50%, and those of MCW0174, ADL0266, ABR0382 were all 31.25%. MSI was found in 30 samples out of 32, the sensitivity of No. 11 sample was up to 45.00%, and that of No. 3 sample was up to 35.00%. Therefore, MSI was found on Chromosome 4 with CEF infected with MDV.
