摘  要:目的观察培养的大鼠皮层神经元损伤后c-fos蛋白表达,探讨其在继发性脑损伤中的作用。方法混合培养10~12天SD胎鼠的皮层神经元分别用谷氨酸和机械划痕损伤。采用SP法进行免疫组化双重标记反应。结果谷氨酸作用后2小时,c-fos蛋白开始表达,4小时后在核中明显浓聚。创伤后2小时仅在创道边缘直接损伤部位有c-fos蛋白表达,远离创道未直接损伤区的神经元在创伤后4小时才出现;少数神经元突起远端有明显c-fos蛋白颗粒;损伤后6小时神经元突起开始崩解,但c-fos蛋白颗粒仍未消失。结论谷氨酸损伤可引起培养的神经元c-fos蛋白的大量表达,损伤后培养皮层神经元和胶质细胞可以释放出神经毒性物质。Aim To observe the expression of c-fos protein in cultured cortical neurons subject to either glutamate or mechanical injury in rats, and to study the effect of c-fos protein on brain injury. Methods Mixed cultured cortical neurons from SD rats were injured by either glutamate or mechanical force, then the expression of c-fos protein was studied with doublelabeled immunohistochemistry. Results Two hours after glutamate injury, the expression of c-fos protein began in the nuclei and 4 hours later it reached peak, and c-fos protein only expressed in the direct injured area at the edge of traumatic tracts after 2 hours and began to express till 4 hours in the indirect injured area far from traumatic tracts. The positive immunoreaction of c-fos protein granule remained. Conclusion The c-fos protein expression can be induced by glutamate. Neurotoxic substances were produced in cultured medium after traumatic injury, and c-fos expression may be associated with neuron apoptosis.
