摘 要:目的:探讨CXCR4在神经母细胞瘤细胞LA-N-5上的表达及其对化疗药物环磷酰胺(CTX)、盐酸吡柔比星(THP)的敏感性,明确化疗药物是否能通过抑制转移相关趋化因子受体的表达发挥作用。方法:体外培养神经母细胞瘤细胞LA-N-5,流式细胞仪和PCR测定LA-N-5细胞上CXCR4蛋白和mRNA的表达。通过MTT试验明确化疗药物CTX和THP对LA-N-5细胞的效应剂量,通过Real-timePCR及流式细胞术在基因及蛋白水平检测化疗药物作用后LA-N-5细胞中CXCR4表达的变化。结果:①流式细胞术检测结果表明CXCR4分子在LA-N-5肿瘤细胞中高表达,两种化疗药物均能使其在蛋白水平表达显著降低,但两种药物之间没有协同作用。②Real-timePCR结果显示化疗药物THP能使肿瘤细胞中CXCR4mRNA的表达降低,而CTX没有此作用,二者在抑制LA-N-5上CXCR4mRNA的表达方面有协同作用。结论:CTX和THP能显著降低转移相关趋化因子受体CXCR4在神经母细胞瘤细胞LA-N-5表面的表达,提示化疗药物发挥作用的机制可能涉及其对趋化因子受体表达的影响。Objective: To observe the expression of CXCR4 on neuroblastoma cells,LA-N-5, and its sensitivity to chemotherapeutic drugs CTX and THP, to investigate whether chemotherapy might affect tumor pathology by inhibiting the expression of this chemokine receptor. Methods: LA-N-5 cells were cultured in vitro. Expression of protein and mRN Aof CXCR4 on LA-N-5 cells were detected by flow cytometry and polymerase chain reaction (PCR). Dose-effects of chemotherapeutic drugs including CTX, THP on LA-N-5 cells were investigated by MTT assay. Then effects of CTX, THP on protein and mRNA expression of CXCR4 on LA-N-5 cells were examined by flow cytometry and real-time PCR assays. Results: CXCR4 expressed on LA-N-5 cells in high levels. Chemothera- peutic drugs CTX, THP decreased the expression of CXCR4 protein on LA-N-5, but no synergistic effect was found between two medicines. At mRNA level, THP but not CTX significantly down-regulated the expression of CXCR4 mRNA, while CTX showed synergistic effects in down-regulating CXCR4 expression by THP. Conclusion: Chemotherapeutic drugs CTX and THP significantly down-regulated the expression of chemokine receptor CXCR4 on LA-N-5 cells, indicating that chemotherapy can inhibit the progression of tumor cells by down-regulating the expression of chemokine receptors.
