摘  要:目的通过改进发明一种新的细菌鞭毛银染色方法。方法本方法通过载玻片的准备、菌种活化、染液制备,菌液制备孵育、制片、染色等环节步骤完成。结果改进后的牛肉膏蛋白胨培养基新配方,营养更丰富,硬度更适宜,镜检结果,鞭毛多,粗壮、着色更容易且均匀。菌液改为恒温水槽孵育,能让菌体鞭毛更活跃,鞭毛更舒展。涂片菌膜清晰便于少量A液覆盖操作,B液染色升温时容易掌握火源高度。结论这种鞭毛染色方法操作简单,易学易掌握,鞭毛着色率高,还有延长A、B液的有效使用时间。此鞭毛染色值得推广使用。Objective To develop a new technique for bactenal flagella staining by impoving the traditional way. Methods It was completed thought the slide was prepared, activation strain, dye preparation, bacteria incubated preparation, production, dyeing and other steps. Results Improved formula of beef extract peptone medium was more nutrition, more suitable hardness. Examination results showed that the flagella were more and thicker and staining more easily and evenly. Incubate the bacterial in Sink temperature could make bacterial flagellum more ac- tive, flagellum more stretch. Film smear was more clear, facilitate coverage of small amount of Reagent A, B Reagent controlled easily temperature when a high degree of fire. Conclusion This method operated easily and easy to be mastered, and the success rate was high. Also to extend the effective use time of reagent A, B. This technique was overcome almost all the difficulties in flagella staining and so could be used as a routine method.
