摘 要:采用DALP分子标记技术,对10个川百合居群进行DNA指纹检测,结果筛选出5个引物组合,扩增后共产生192条DNA基因片段,其中178条谱带具有遗传多态性,占92.71%;10个居群的总等位基因数(Na)为1.9271、平均为1.0823,总有效等位基因数(Ne)为1.4430、平均1.0533,基因多样性指数(H)为0.2673、平均0.0303,总Shannon多样性指数(I)为0.4105、平均为0.0447,基因分化系数(Gst)为0.8874,即遗传变异有88.74%发生在居群间、有11.26%发生在居群内,表明川百合居群间存在较大的遗传分化。此外,昆明市西山的川百合野生居群(DC)的遗传多样性明显高于栽培居群,这与川百合栽培品种长期人工驯化有直接关系。同时野生居群受到人为活动干扰严重。研究结果表明,DALP分析可为川百合遗传育种的可持续利用提供理论依据。DALP(Direct amplification of length polymorphism)was applied to detect genetic polymorphism of 10 populations of Lilium davidii Duchartre.Five primer groups were screened, and a total of 192 DNA fragments were amplified, among which 178 were polymorphic, i.e. the percentage of polymorphie bands (PPB) was 92.71%. The total observed number of alleles (Na) wasl.9271 and the average Na wasl.0823; the total effective number of alleles (Ne) was 1.4430 and the average Ne was 1.0533; the total Nei's gene diversity(H) was 0.2673 and the average H wasO.0303; the total Shannon's Information index (I) was 0.4105 and the average I was 0.0447. The coefficient of gene differentiation (Gst) was 0.8874 between populations, namely 88.74% genetic variation occurring between populations and rest 11.26% within population,the result indicated that L. davidii had much larger genetic differentiation among populations.The genetic diversity of population in Xishan Kunming (DC)was evidently higher than that planting populations, this was directly related to the long acclimation of L. davidii by artificial culture, and wild populations were severely interfered by human activities. DALP analysis provides this species with molecule evidence for the sustainable exploitation of genetic breeding.
