摘 要:[目的]为探索MS2噬菌体的宿主菌范围及在不同的宿主菌内的生长情况等,为流行病学研究、细菌的噬菌体分类鉴定和其他相关研究提供基本数据和资料。[方法]采用双层琼脂法测定大肠埃希菌MS2噬菌体对各试验菌株的裂解情况,观察其噬菌斑生长情况并计数。[结果]①大肠埃希菌MS2噬菌体与埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B分别进行双层琼脂平板培养,结果均出现大小均一、边缘整齐、透明清晰的噬菌斑;②相同稀释度的MS2噬菌体与埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B培养产生的噬菌斑数量依次递减;③MS2噬菌体与其余6种所选试验菌株分别进行双层琼脂平板培养,结果琼脂表面均出现浑浊,未产生噬菌斑。[结论]在本次试验所选的8种试验菌中,通过双层琼脂法测定适合做大肠埃希菌MS2噬菌体宿主菌者有埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B;所选其余6种试验菌株均不适合做MS2噬菌体的宿主菌。[Objective] To search the host range of the MS2 phage and its vegetative status in some correlated hosts, and to provide the basic data for epidemiologic stud), bacterial identification, and other studies. [Methods] We assessed the cleavage of the MS2 phage for each test organism through the double-agar-layer plaque technique. Meanwhile, we observed the status and quantity of the plaque and selected the hosts of the MS2 phage among test organisms. [ Results ] ① The plaques, uniform, regular and limpid were tested by the double-agar-layer plaque technique with the activity of the MS2 phage and E-coil ATCC 15597, E-coil 285, E-toll B, respectively. ②The number of the plaque generated by the MS2 phage and E-coil ATCC 15597, E-coil 285, E-coil B respectively decreased one by one. ③ The result of the MS2 phage and other six test organisms all showed some bacterias on the surface but no plaque. [ Conclusion] This study shows the hosts of the MS2 phage are E-coil ATCC 15597, E-colt 285 and E-coil B among the test prganisins by double-agar-layer plaque technique, and the other six tested trains are unfit for the hosts of MS2 phage.
