摘  要:[目的]探讨口服血管紧张素II1型受体阻滞剂(angiotensin II type 1 receptor blockade,ARB)奥美沙坦(olmesartan)对自发性高血压大鼠(spontaneously hypertensiverats,SHR)延髓腹外侧头端(rostral ventrolateral medulla,RVLM)局部肾素血管紧张素系统(rennin-angiotensin system,RAS)的影响。[方法]雄性12周龄SHR14只,随机分成SHR组(正常饮水,n=7)及SHR-Olme组(olmesartan,10μg/(g.day),n=7),WKY组为正常血压大鼠7只(olmesartan,10μg/(g.day),n=7)。喂养4周后在延髓腹外侧头端微量注射血管紧张素II(angiotensinII,AngII),观察平均动脉压(meanarterialpressure,MAP)和心率(heartrate,HR)的变化。[结果]AngII可使SHR组MAP明显升高(+14.1±4.6mmHg),SHR-Olme组轻度升高(+3.7±1.4mmHg,),2组比较差异有统计学意义(P﹤0.05),WKY组MAP升高不明显(+2.0±0.8mmHg),与SHR组及SHR-Olme组比较,差异有统计学意义(P﹤0.05);3组心率变化差异无统计学意义。[结论]奥美沙坦能通过影响脑组织局部肾素血管紧张素系统产生降压作用。[Objective] To investigate the central effect of oral administration of olmesartan (art AT1 receptor antagonist) on the rennin-angiotensin system in the Rostral ventrolateral medulla of spontaneously hypertensive rats. [ Methods] 14 male SHRs were randomly divided into SHR group (normal water, n=7) and SHR-Olme group (olmesartan in drinking water, 10μg/(g.day), n=7), age-matched wistar-Kyotorats (WKY) (olmesartan in drinking water, 10μg/(g.day), n=7).After fed for 4 weeks, microinject angiotensin II (AngII) in the rostral ventrolateral medulla, the changes of the mean arterial pressure and heart rate were observed. [ Results ] The pressor responses to microinjeetion of AngII in the Ohnesartantreated SHR (+3.7±1.4mmHg) were significantly weaker than those in untreated SHR (+14.1±4.6mmHg); however, they were still greater than those in the Wistar-Kyoto rats (+2.0±0.8mmHg, P 〈 0.05); the changes in HR did not differ among the three groups. [Conclusion ] Olmesartan may partly normalize the arterial pressure by acting on the central rennin-angiotensin system.
