二硫化碳经皮染毒对小鼠骨髓细胞及精子影响作用的研究

摘  要:目的研究二硫化碳(CS2)经皮染毒对小鼠骨髓细胞及精子细胞的毒性作用。方法选择无特异性病原体级健康成年雄性NIH小鼠50只,采用完全随机的方式分为5个组,分别为:CS2高剂量组[900mg/kg,即1/2半数致死浓度(LD50)]、中剂量组(360mg/kg,即1/5LD50)、低剂量组(180mg/kg,即1/10LD50),阴性对照组(生理氯化钠)和阳性对照组(环磷酰胺150mg/kg),采用封闭涂皮法染毒,每天1次,每周5d,直至实验结束。于第6天处死部分小鼠进行骨髓细胞微核检测,于第35天处死其余小鼠进行精子畸形实验,显微镜下观察并计数微核及畸形精子数。结果CS2中、高剂量组骨髓细胞微核数高于阴性对照组,差异具有统计学意义(P<0.05),同时低、中、高剂量组精子畸形率高于阴性对照组,差异具有统计学意义(P<0.05)。结论一定剂量的CS2经皮染毒对雄性小鼠的骨髓细胞及精子细胞具有一定的损伤作用,提示CS可能具有致突变作用。Objective To study the mutagenicity of carbon disulfide (CS2 ) in mice by administered through skin. Methods 50 healthy male NIH mice were randomly divided into 5 groups: CS2 high-dose group (900 mg/kg) ,medium-dose group (360 mg/kg) ,low-dose group (180 mg/kg) ,negative control group (NS) and positive control group (cyclophosphamide, 150 mg/kg). All the mice were treated with the respective solution per eutem ,once working day for 5 consecutive weeks. On the sixth day, half mice of each group were sacrificed to test the marrow micronucleus rate. The other mice were sacrificed to assay sperm abnormality on the 35th day. Results There were significant increases of the frequencies of micronuclei in medium and high dose groups compared with the negative control group( P 〈 0. 05 ). The frequencies of the abnormal sperm also increased signifieantly(P 〈0. 05) in all CS2 groups as compared with the control. Conclusion The results showed that CS2 might be a mutagene in mice when absorbed through skin.
