摘  要:目的观察四物汤配方颗粒对骨髓抑制小鼠造血功能的影响,探讨其对造血微环境调控作用机制。方法50只小鼠随机分成5组:正常组、模型组、3个不同剂量给药组。观察各组骨髓有核细胞(BMNC)对骨髓基质细胞(BM-SC)的黏附能力,应用免疫印迹法(WB)测定四物汤配方颗粒对骨髓抑制小鼠BMSC层粘连蛋白(LN)、纤维粘连蛋白(FN)、焦点黏附激酶(FAK)的影响。结果四物汤配方颗粒各组均能明显促进LN、FN、FAK表达;并具有剂量依赖性,表现出高剂量组BMSC对正常骨髓细胞的黏附能力最明显、对LN、FN、FAK表达最高。结论四物汤配方颗粒可通过对骨髓抑制小鼠造血微环境的改善,促进骨髓造血功能的恢复。Objective To investigate the effect of formula granule of Siwu abstract on hematogenesis function in bone marrow depression mice and to explore its mechanism of hematopoietic microenvironment regulation. Methods 50 mice were randomly divided into 5 groups, a normal group, a control group and 3 dose groups. The adhesion between bone marrow stromal cell (BMSC) and bone marrow nucleated celI(BMNC) was observed,and the effects of the formula granule on Laminin(LN) ,fibronectin(FN) ,focal adhesion kinase(FAK) of the BMSC in bone marrow depression mice were explored by western blotting (WB). Results It was found that the expressions of LN, FN, LAK were obviously increased in all dose groups. Conclusion The results suggested that formula granule of Siwu may improve hematopoietic microenvironment and restore the hematopoietic function in bone marrow depression mice.
