芦笋试管培养诱导时间和培养基成份对试管苗生根的影响

摘　　要：在芦笋试管苗诱导根原基（生根的第一阶段）时，添加不同种类和浓度激素的诱导培养基，诱导生根的效果不同，且各有一个最适的诱导时间，在这个时间内，生根率为最高，短于或长于这一时间，生根率会随之减少。其中１／２ＭＳ＋ＮＡＡ０．１ｍｇ／ｌ＋ＩＢＡ０．５ｍｇ／ｌ＋ＫＴ０．１ｍｇ／ｌ＋ＰＰ３３３２ｍｇ／ｌ＋ＶＢ１０．４ｍｇ／ｌ＋蔗糖２％＋琼脂０．６％诱导培养基，诱导生根的效果较好，并以诱导１１天的生根率最高，达８７．１％；每株根长以诱导５天最长，为５．０５ｃｍ；每株根数以诱导１３天的最多，达４．３６条。诱导生根率最低的是１／２ＭＳ＋ＮＡＡ０．５ｍｇ／ｌ＋ＫＴ０．０５ｍｇ／ｌ＋ＶＢ１０．１ｍｇ／ｌ＋蔗糖２％＋琼脂０．６％培养基，诱导１３天，仅为２３．０％。The explants of asparagus (cv.32X228) were placed in 6 introducing culture media to initialize the roots for 3 to 15 days.Then they were transplanted in the rooting media for 30 days.Results showed that the explants grown in the introducing culture medium containing 1/2 MS+NAA 01mg/l+IBA 05mg/l+KT 01mg/l+PP333 2mg/l+VB1 04mg/l+sugar 2%+agar 0.6% for 11days had the highest rooting rate of 871%.Root length was highest(5.05cm) in the treatment for 5 days.Root number (436)was highest in the treatment for13 days.The rooting rate (23%)was lowest in the treatment of 1/2 MS+NAA 05mg/l+KT 005mg/l+VB101mg/l+sugar 2%+agar 06% for 13 days.
