摘 要:根据已发表的PRRSVIAF-exp91株的核酸序列,设计了一对特异性引物,用RT-PCR扩增了PRRSVCH-la株的N基因,经补平和磷酸化后,克隆到pUC18SmaI位点上。经电泳分析、酶切和PCR鉴定证实后,进行序列测定。序列分析表明CH-la株与IAF-exp91株(美洲型)N基因核苷酸同一性为93.2%,氨基酸同一性高达96.7%,而与LV株(欧洲株)核苷酸同一性为63%,氨基酸同一性仅为59%。其推导氨基酸残基分子量为13.3kDa。根据CH-la株N基因推导的氨基酸残基缺少欧洲型毒株LV特有的两个氨基酸片段,表明CH-1a株与IAF-exp91株同源性高于它与LV株的同源性,而且CH-1a株与鼠乳糖脱氢酶增高症病毒(LDV)在进化关系上比LV株与LDV更密切。CH-1a株N基因推导的氨基酸残基N-端1/2段约26%是由Arg、Lys和His这3种氨基酸组成的,其疏水性侧像图与LV株和VR-2332株(美洲型)几乎相同,这表明两种基因型的核衣壳蛋白(N蛋白)具有一定的保守性。由于N蛋白是PRRSV所有结构蛋白中免疫原性比较强的病毒蛋白,是现行血清学试验的抗原基础,因此N基因的分子克隆为研制PRR?Porcine reproductive and respiratory syndrome virus (PRRSV) is the causative agent of porcine reproductive and respiratory syndrome (PRRS),a newly recognized pig disease of economic importancePRRSV strain CHla was recently isolated in a pig farm in China during an outbreak of PRRS,and it was shown to be American genotype in an earlier studyA fragment (encompassing fulllength of N gene)amplified by RTPCR was cloned into SmaI site of plasmid pUC18,and a recombinant harboring the N gene was designated as pUC18ORF7The purified recombinant was subjected to sequencingThe N gene of CHla displayed 932% nucleotide identities and 967% amino acid identities with strain IAFexp91(a reference strain of Canada),European gerotype),and only 63% nucleotide idetities and 59% amino acid identities with strain LV(Euroqean gerotype)The deduced amino acids of the N gene have a molecular weight of 133 kDa,and six hydrophobic regions were identifiedThe hydrophobicity profiles are nearly identical between CHla and LV or VR2332(American genotype)The CHla N protein,however,was found to be shorter than that of LV by five amino acids The results indicate that CH1a is closely related to IAFexp91,but not so to LV. As N gene of PRRSV is relatively conservative,the cloned N gene in the study will be expressed in further as antigen for diagnostic use,as well as possible vaccine candidate.
