摘 要:研究【目的】检测秦川牛CS-1基因的单核苷酸多态性(SNPs),并将其与秦川牛胴体和肉质性状进行关联分析。【方法】采用PCR-SSCP方法对485头秦川牛CS-1基因第3外显子进行多态性检测,分析其与部分胴体、肉质性状的关系;【结果】在CS-1基因第3外显子191碱基处发现C→A突变,201碱基处发现C→T突变。检测到A和C两个等位基因,其基因频率分别位0.1381和0.8619。方差分析结果表明:胴体性状方面,AC型个体背部皮下脂肪厚显著高于CC型个体(P<0.05);在肉质性状方面,AC型个体的大理石花纹显著高于CC型个体(P<0.05),AA型和AC型个体的嫩度要显著优于CC型个体(P<0.05);其余性状差异不显著;【结论】揭示可以用该多态性位点(CS-1 Exon3)对秦川牛的胴体性状和肉质性状进行标记辅助选择。[OBJECTIVE]This research was to detect the polymorphism in exon 3 of CS-lgene and its relationship with some carcass and meat quality traits of Qinchuan eattle.[METHOD]PCR-SSCP and DNA sequencing methods were employed to reveal the polymorphism of exon 3 and its flanking within CS-1 gene in 485 Qinchuan cattle, and analysing its relationship with some carcass and meat quality traits of Qinehuan cattle;[RESULTS]C→A mutation in 191bp and C→T mutation in 201bp of CS-1 gene exon 3 were found. There were two allele gene A and C in this locus with the corresponding frequencies of 0.1381 and 0.8619. Statistical analy- sis indicated that individuals with genotype AC had superior thickness of back fat and marbling (P〈0.05) when compared those with genotype CC; As for the tenderness, animals of AA and AC genotype were more excellent than animals with genotype CC(P〈0.05);[CONCLUSION]It suggested that this locus could be used as a molecular marker in Qinchuan cattle breeding for carcass and meat traits.
