摘 要:[目的]优化荷叶总黄酮的提取工艺条件。[方法]以安徽合肥地区产的风干荷叶为材料,用乙醇热提法提取荷叶黄酮。以影响提取得率的因素固液比、提取时间、提取温度和乙醇体积分数为因素,通过正交试验筛选提取荷叶总黄酮的最佳工艺条件,并通过单因素试验对提取条件的重现性与稳定性进行验证。[结果]由正交试验极差K值可知各因素对荷叶黄酮提取得率影响的顺序为:提取温度>固液比>乙醇体积分数>提取时间;根据节能的角度最终得出提取荷叶总黄酮的最佳工艺条件是:提取温度为80℃、料液比为1∶20、乙醇体积分数为50%、提取时间为3 h。单因素试验表明,重现性与稳定性试验与正交试验所得的结果基本符合。[结论]该研究为荷叶黄酮开发利用提供一定的参考依据。[ Objective ] The study aimed to optimize the technology conditions of extracting flavonoids from lotus leaf. [ Method ] With air - dried lotus leaf produced from Hefei area in Anhi Province, its flavonoids was exacted by hot extraction with alcohol. With the ratio of solid to liquid, extraction time and temperature and alcohol concn, as the factors that effected the extraction yield, the technology conditions of extracting flavonoids from lotus leaf were screened through the orthogonal test and the reproducibility and stability of the extraction conditions were validated through single factor test. [ Result ] Based on the range K value of orthogonal test, the effect of each factor on the extraction yield of flavonoids from lotus leaf in order was extraction temperature 〉 ratio of solid to liquid 〉 alcohol conch. 〉 extraction time. The optimum technology conditions of extracting flavonoids from lotus leaf from the view of saving energy were as follows : extraction temperature of 80 ℃, ratio of solid to liquid of 1:20, alcohol concn, of 50% and extraction time of 3 h. The single factor test showed that the results from the reproducibility and stability test accorded with that from the orthogonal test. [ Conclusion ] The study provided some refernce basis for the develplment and utilization of the flavonoids from lotus leaf.
