摘　　要：目的观察DC-Chol脂质体DNA复合物(DLDC)体外抗乙型肝炎病毒(HBV)的作用。方法制备DLDC,HepG2.2.15细胞经不同浓度的DLDC作用后,检测其对细胞的毒性作用;提取细胞质DNA,用化学发光Southern blot法检测DLDC对细胞内HBV DNA复制的抑制作用。结果DLDC可抑制HepG2.2.15细胞增殖,其TC0为78.13 ng/ml,TC50为421.86 ng/ml;DLDC可抑制HepG2.2.15细胞HBV DNA复制,其IC50为45 ng/ml,SI为9.37。结论DLDC对胞内HBV DNA的复制有明显的抑制作用。Objective To observe the inhibitory effect of DC-Chol liposome DNA complex （DLDC） on HBV in vitro. Methods HepG2.2.15 cells were treated with prepared DLDC at various concentrations, and the toxicity of DLDC to the cells was determined by MTT method. The cytoplasmic DNA of treated HepG2.2.15 cells was extracted, and the inhibitory effect of DLDC on replication of HBV DNA in cells was determined by chemiluminometry and Southern blot. Results The TC0 and TC50 of DLDC to HepG2.2.15 cells were 78. 13 and 421. 86 ng/ml, while the IC50 and SI to HBV DNA were 45 ng/ml and 9. 37, respectively. Conclusion DLDC showed significantly inhibitory effect on HBV DNA replication in HepG2.2.15 cells.
