摘 要:目的:研究中药复方对大脑中动脉闭塞(MCAO)模型大鼠脑内Nogo-A的影响,探讨其促进神经发生的作用。方法:30只大鼠随机分为药物组(治疗组)、病理模型组(对照组)、正常对照组(假手术组)。用Tamura法造成MCAO大鼠模型,于造模成功后6天药物组灌胃给予中药复方首乌仙海片10g·kg-1,余两组分别灌胃给予同等量生理盐水,日1次,共2周。用原位杂交法观察MCAO大鼠脑内Nogo-A的表达。结果:药物组大鼠脑内Nogo-A表达明显减弱。结论:中药复方首乌仙海片可显著抑制MCAO大鼠脑内Nogo-A的表达,从而促进缺血性卒中神经发生。Objective:To study the effect of complex of Chinese herb on the expression of Nogo protein-A in the brain of MCAO rats and explore the action mechanism of its promoting neurogenesis.Methods:30 rats were randomly divided into drug group,model group and normal control.The models of MCAO rats were successfully established by the method of Tamura.After six days,the rats of drug group had been taken orally Shouwuxianhai tablets by 10g/kg crude drug one time daily for two weeks,and the rest groups had been taken orally equivalence saline once a day for two weeks.The expression of Nogo protein-A in the brain of MCAO rats was observed in situ hybridization.Results:The expression of Nogo protein-A in the brain of MCAO rats in drug group was obviously decreased.Conclusions:Shouwuxianhai tablets can predominantly inhibit the expression of Nogo protein-A in the brain of MCAO rats.Therefore,it can promote the neurogenesis of AICD.
