发酵支原体、梨支原体及穿通支原体致死性感染大鼠血清细胞因子检测

摘  要:目的:探讨发酵支原体(Mycoplasma fermentans,Mf)、梨支原体(Mycoplasma pirum,Mpi)及穿通支原体(Myco-plasma penetrans,Mpe)致死性感染免疫缺陷大鼠血清TNF-α、IL-1β、IL-6和IL-10含量与重要器官损伤之间关系。方法:清洁级SD大鼠61只,分免疫抑制组(47只)、非免疫抑制组(7只)和生理盐水对照组(7只)。免疫抑制组预先经环磷酰胺隔天注射(50 mg/kg.d)3次制备免疫抑制大鼠模型,再分别腹腔注射0.6 ml 3种支原体标准菌株Mf、Mpi、Mpe以及本实验室分离株(W12)和无菌生理盐水;非免疫抑制组和生理盐水对照组取正常大鼠腹腔分别注射相同剂量Mf和生理盐水。取大鼠的肝、肾和脑组织做电镜观察,同时分别取大鼠血液采用ELISA方法检测其肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)和白细胞介素10(IL-10)的含量变化。结果:Mf感染组大鼠血清TNF-α、IL-1β和IL-6的含量明显升高(P<0.01);免疫抑制3种支原体感染组血清TNF-α、IL-1β和IL-6的含量均明显低于非免疫抑制Mf感染组(P<0.01),而IL-10含量却显著高于非免疫抑制Mf感染组(P<0.01)。电镜结果显示大鼠的肝、肾和脑组织内细胞间质高度水肿、线粒体肿胀、器官实质变性坏死。结论:发酵支原体、梨支原体以及穿通支原体对免疫缺陷机体产生进一步的细胞免疫抑制作用,更易使免疫缺陷机体发生多器官衰竭甚至致死性感染。Objective:To study changes of serum cytokines in rats with the fatal infection changed by mycoplasma fermentans (Mf), mycoplasma pirum(Mpi) and mycoplasma penetrans (Mpe). Methods:61 clean SD rats which were and nonimmunoanppression rats group. The former (47 rats) were prepared immunosuppression rats mode by injecting intraperitoneally with cyclophosphamide 50 mg/kg/d every other day and were received 0. 6 ml Mf, Mpi, Mpe, this experiment isolation strain(W12 ) and sterile physiological salt solution by intraperitoneal injection. The latter (14 normal rats) were injected with Mf and sterile physiological salt solution in the same dose as control group. The changes of ultrastructure of liver, kidney and brain were observed and the concentration of serum TNF -α, IL - 1 β, IL -6 and IL - 10 were also detected with enzyme -linked immunosurbent assay (ELISA) in rasts. Results:The level of TNF- α, IL- 1 β and IL- 6 in rats infected by Mf was significantly higher than those of normal rats (P 〈0. 01 ) ; The level of TNF - α, IL - 1β and IL -6 in the immunosuppression group rats infected with three Mycoplasmas was significantly lower than those of the nonimmunosuppression group rats ( P 〈0. 01 ), however, The level of IL - 10 was significantly higher than that of the nonimmunosuppression group rats ( P 〈0. 01 ). Ultrastructural observation showed that there were appeared the inflammatory reaction, including interstitial tissues cell dropsy, cell mitochondrial crista swollen, vacuole changes occurred and multiple organ damaged in liver, kidney and brain of the dead rats. Conclusion :The infection of Mf, Mpi and Mpe could lead to further immune suppression and multiple organ failure in the immunodeficiency body.
