二氧代哌嗪苯磺酰环己脲降糖作用的研究

摘  要:目的:研究观察二氧代哌嗪苯磺酰环己脲对链脲佐菌素(STZ)诱导的2型糖尿病小鼠血糖血脂的影响以及刺激SMMC7721细胞对3H-2-脱氧葡萄糖吸收影响。方法:以高脂高热量饲料喂养及腹腔注射链脲佐菌素复制小鼠2型糖尿病模型,采用酶比色法测定糖尿病小鼠的空腹血糖、甘油三酯(triglyceride,TG)、胆固醇(cholesterol,CHO)等相关生化指标;利用液闪计数法检测细胞对3H-2-脱氧葡萄糖的吸收。结果:二氧代哌嗪苯磺酰环己脲可明显降低血糖水平,改善甘油三酯、胆固醇(P<0.01);与对照组比较,二氧代哌嗪苯磺酰环己脲可显著增加SMMC7721细胞对葡萄糖的吸收(P<0.01),3H-2-脱氧葡萄糖的吸收值增加近2倍,并且显著增强胰岛素刺激的葡萄糖吸收作用。结论:二氧代哌嗪苯磺酰环己脲有改善2型糖尿病小鼠血糖血脂的作用;并且具有刺激葡萄糖吸收以及增加胰岛素刺激的葡萄糖吸收作用。Objective : To explore the effects of 1 -(4 - [ 2 -( 1 - ethyl- 2,3 - dioxopiperazine4-carboxamido )ethyl ]-phenylsulfonyl)-3 -eyclohexyl urea on the fasting blood glucose and the blood lipemia in the streptozotocin (STZ)-induced diabetic mice and the glucose uptake in SMMC7721 cells. Methods:The animal models of Type 2 diabetes were prepared by nourishment of high calorie and high fat and by injection of STZ. The enzyme colorimetry was used to detect the level of fasting blood glucose,triglyceride ( TG), cholesterol (CHO) and scintillation was employed to examine the glucose uptake in SMM7721 cells. Results:The drug effectively decreased the blood glucose and improved the metabolism of blood lipemia in the STZ -induced mice. Compared with the control cells, the drug significantly improved the glucose uptake in SMMC7721 cells (P 〈 0.01 ) and the uptake of 2-deoxy-[ 3 H ] glucose in SMMC7721 cells increased by twice, with evident insulin-stimulated glucose uptake (P 〈 0.01 ). Conclusion:The new drug could modify the blood glucose and lipemia in STZ- induced mice and improve the glucose uptake in SMMC7721 cells,and it may be a potential anti-hyperglycemic agent.
