摘  要:目的:考察乙酰丹参素单硝酸异山梨醇酯(AcDI-302)对心肌缺血/再灌注(MI/R)大鼠的心肌保护作用,并与单硝酸异山梨醇酯(ISMN)进行比较.方法:常规制备大鼠MI/R(30 min/3 h)模型.随机分为假手术组,MI/R组,(MI/R+ISMN)组,(MI/R+AcDI-302)组.观察各组大鼠缺血30 min再灌注3 h心功能恢复情况和3 h末血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)活性和丙二醛(MDA)的水平及NO的含量.结果:MI/R+AcDI-302组大鼠较ISMN组心肌梗死范围明显减小,血清中CK,LDH活性和MDA浓度也显著降低,左室收缩压(LVSP)、左室等容收缩/舒张期压力上升或下降最大速率(±dp/dtmax)显著提高.结论:AcDI-302对心肌缺血再灌注损伤大鼠的心肌有保护作用,作用强于ISMN.AIM: To study the protective effect of a new compound, Aeetyl Danshen Acid Isosorbide Mononitrate (AcDI-302), on myocardial ischemia/reperfusion(MI/R) injury and to compare its effect with that of Isosorbide Mononitrate (ISMN). IVIETHODS: Male Sprague-Dawley rats were subjected to 30min of myocardial isehemia and 3 h of reperfusion. The rats were randomly assigned to SHAM group, treatment group of MI/R + solvent and MI/R + ISMN and treatment group of MI/R + AeDI-302. MI/R models were prepared by temporary occlusion of the left anterior descending coronary artery. Heart rate (HR), left ventricle maximum systolic pressures (LVSP) and the maximal first derivative of developed pressure(±dp/dtmax) were monitored throughout the experiments. Myocardial infarction (MI) size and serum ereatine kinase (CK) , lactate dehydrogenase (LDH) , nitric oxide(NO) and malondialdehyde(MDA) of the rats were deter- mined at the end of reperfusion. RESULTS: Compared with ISMN, the new compound of AcDI-302 improved the cardiac function by increasing LVSP and ±dp/dtmax, and decreased the size of myocardial infarction, the activities of CK and LDH and the level of MDA. CONCLUSION: Compared with the protective effect of ISMN, the new compound of AcDI-302 has stronger protective effects on MI/R injury in rats.
