摘 要:背景溃疡性结肠炎发病率呈逐年上升趋势,但发病机制不明,众多基于基因水平的研究不能完全阐明蛋白质水平的变化,差异蛋白质组学是从蛋白质水平研究溃疡性结肠炎发病机制的新方法。目的运用蛋白组学方法探讨溃疡性结肠炎的发病机制。方法选取12例活动期溃疡性结肠炎患者及12例正常肠黏膜为研究对象,提取组织蛋白做2-D电泳,银染,胶图分析找差异点,质谱鉴定蛋白质,最后Western-blot验证差异点。结果差异蛋白组学共鉴定出蛋白质30个(上调16个,下调14个),成功选取其中的vimentin,并得到Western-blot方法的验证。结论溃疡性结肠炎患者肠黏膜存在vimentin降解片段蛋白水平的高表达,这种改变可能在在溃疡性结肠炎的发病机制中起重要作用。Background The incidence of ulcerative colitis (UC)is increasing, but the pathogenesis of UC remains unclear. Comparative proteomic analysis, a new strategy at the protein level, has been used to detect the pathogenesis of UC. Aims To investigate pathogenesis of UC by comparative proteomic analysis. Methods Colonic tissue samples of 12 UC patients in active stage and 12 normal control were taken. Protein separation and identification were performed by 2DE and MALDI TOF/TOF MS. Vimentin degradation frag- ments and protein expression were measured using Western-blot. Results A total of unique 30 different proteins were identified by comparative proteomic analysis. Among them, 16 proteins were up-regulated in UC, 14 proteins were down-regulated. Vimentin degradation fragments were deteced in UC musoca using Western-blot. The expression level of vimentin fragments in UC was noticeably higher then in control. Conclusion There was an increased vimentin degradation in ulcerative colitis, which might play an important role in the pathogenesis of UC.
