pTAT-HA质粒载体在受体菌DH5α中的转化及鉴定

摘　　要：目的探索pTAT-HA质粒在受体菌DH5α中转化的方法。方法配制LB培养基,将pTAT-HA质粒转化至DH5α,培养筛选成功转化子,电泳鉴定。结果pTAT-HA质粒转化菌可在Amp+LB培养基中生长,电泳结果显示质粒大小准确无误(约3000bp)。结论pTAT-HA质粒转化成功,转化菌可冷冻保存质粒,提取的质粒可用于下游分子生物学实验。Objective To explore the method that DH5α was transformed with pTAT-HA vector. Methods LB medium was prepared, pTAT-XIAP vector was transformed into E. coli. DH5c~. competent cells. Results Transformants were cultured on LB medium and selected. The plasmids were extracted,purified and i- dentified by agarose gel electrophoresis. Transformed DH5c~ Could grow on LB medium containing ampicillin, gel electrophoresis analysis showed that the molecular weight of pTAT-XIAP vector was about 3000 bp consistent with the vector. Conclusion The pTAT-XIAP vector was succesfully transformed into the DH5α and the trans- formants were stored by cryopreservation, the purified plasmid could be used for molecular biologic experiments in the future.
