摘  要:针对当前对生物酶降解淀粉作用机理研究不足的问题,通过DNS比色法,考察了不同淀粉生物酶对淀粉的降解过程,并考察了温度、pH值和表面活性剂等外界条件对生物酶降解性能的影响。研究发现,pH值对淀粉酶的降解能力影响最大,在pH值为6.0时其活性最高,而在pH值为8时效果最差;温度对淀粉酶的影响也较大,温度从50℃升高到75℃后,淀粉酶的活性对pH值变得更加敏感,碱性环境对酶解的抑制作用更强;表面活性剂对淀粉酶具有抑制作用,且其浓度越大,抑制作用越强。该研究结果有助于指导淀粉酶的现场应用。DNS colorimetry method is used to study the effect of amylase on the degradation of starches in different conditions such as temperature,pH value and the presence of surfactants.It is found that pH value has the greatest impact on the degradation efficiency of amylase.Amylase has the highest activity at pH 6.0 and the lowest activity at pH 8.0.Temperature also has a relatively strong influence on the performance of amylase.When temperature is increased from 50 ℃ to 75 ℃,the activity of amylase becomes more sensitive to the change of pH value,and high pH has a much stronger inhibition to the performance of amylase.Surfactants inhibit the performance of amylase,the higher the concentration of a surfactant,the stronger the inhibition to the performance of amylase.
