脂欣康干预泡沫细胞内胆固醇及胆固醇酯分析

摘  要:目的观察脂欣康干预大鼠血管平滑肌细胞(VSMC)源性泡沫细胞胆固醇和胆固醇酯含量的变化,以探讨脂欣康对动脉粥样硬化(As)泡沫化细胞形成的可能调控机制。方法用组织块培养法培养VSMC,以氧化低密度脂蛋白使其泡沫化,并以脂欣康、洛伐他汀分别干预。进行高效液相色谱分析,分离纯化胆固醇和胆固醇酯,观察其含量变化。结果脂欣康与洛伐他汀均能降低总胆固醇及胆固醇酯含量,并且胆固醇酯与总胆固醇的比值均降至50%以下,与生理盐水组、空白对照组相比均有统计学意义(P<0.01)。结论脂欣康显著抑制VSMC源性泡沫细胞胆固醇和胆固醇酯的增高,调脂作用是其抑制VSMC泡沫化和抗As的机制之一。Objective To observe the effect of Zhixinkang(ZXK) on cholesterol and cholesteryl ester in vascular smooth muscle cells(VSMCs)-derived foam cells. Methods The VSMCs were treated by ZXK and lovastatin. The expression of cholesterol and cholesteryl ester were measured by high performance liquid chromatography(HPLC). Results The espression of Cholesterol and cholesteryl ester in ZXK and Lovastatin groups were lower than that in isotonic na chloride group and the blank group(P〈0.01 ). The ratio between cholesterol and cholesteryl ester in ZXK and lovastatin groups droped below 50 %. Conclusion ZXK could decrease cho lesterol and cholesteryl ester in VSMCs - derived foam cells. That could be one of mechanism to restrain the formation of VSMCs foam cell.
