益气活血方对球囊损伤大鼠颈总动脉胶原的影响

摘　　要：目的探讨益气活血方对球囊损伤大鼠颈总动脉增殖的干预机制,对损伤血管壁Ⅰ型、Ⅲ型胶原和基质金属蛋白酶2(MMP2)的表达进行观察。方法48只雄性SD大鼠随机分为对照组,模型组,益气活血方大、小剂量组,球囊损伤前1d给对照组与模型组生理盐水灌胃,益气活血方组给予中药灌胃,至第5天和第28天处死大鼠,取其损伤的颈总动脉,用免疫组织化学方法检测两个时相点MMP2蛋白和第28天Ⅰ型胶原、Ⅲ型胶原的表达。结果对照组I型胶原及MMP2表达阳性,对照组Ⅲ型胶原表达阴性或有极少量表达,模型组各指标表达明显增多,用药后各组表达显著减少,与模型组比较有统计学意义(P<0.01);益气活血方大、小剂量组之间比较无统计学意义(P>0.05)。结论益气活血方对球囊损伤后血管增生的抑制作用机制与下调I型胶原、Ⅲ型胶原及MMP2的表达,抑制血管细胞外基质,进一步减少血管重构有关。Objective To investigate the effect of Yiqihuoxue decoction on expression of collagen Ⅰ ,collagen m and matrix metal- loproteinase- 2 （MMP2）after carotid artery vascular injury in rats. Methods Forty - eight rats were randomly divided into four groups：Control group, model group, Yiqihuoxue decoction low dose group and Yiqihuoxue decoction high dose group. The balloon catheter of 2F Fogarty was performed on the left common carotid artery. The expression of the MMP2 was observed at 5th and 28th day,and the expression of collagen Ⅰ ,collagen Ⅲ were observed at 28th day after injury by immunohistochemical technique. Results Compared with control group,the expression of collagen Ⅰ,collagen Ⅲ and MMP2 were increased significantly in model group. Compared with model group,the expression of collagen Ⅰ,collagen Ⅲ and MMP2 were decreased significantly in Yiqihuoxue decoction low dose group and Yiqihuoxue decoction bigh dose group（P〈0.01 ）. There was no statistical significance in Yiqihuoxue decoction low dose group and Yiqihuoxue decoction high dose group（P〉0.05）. Conclusion Yiqihuoxue decoction could decrease the vascular proliferation after balloon injury, which might be associatcd with inhibiting the proliferation of extracellular matrix and vascular smooth muscle cell through reducing the expression of collagen Ⅰ,collagen m and MMP2.
