摘 要:目的:观察γ射线辐照外周血单个核细胞对培养人前列腺癌细胞系PC-3细胞的杀伤作用。方法:实验设PC-3P肿瘤细胞对照组、单纯外周血单个核细胞对照组、照射和未照射的外周血单个核细胞与肿瘤细胞共培养组,照射剂量为1Gy,吸收剂量率为17Gy/min。利用吖啶橙/溴化乙锭(AO/EB)荧光双染色法和MTT比色法,观察外周血单个核细胞对PC-3细胞的杀伤情况。结果:外周血单个核细胞在照射后体外培养至144h有大量细胞死亡,存活细胞明显少于未照射组,培养至240h时,照射与非照射组中存活的外周血单个核细胞均减少,但是在照射的各组中减少更为明显。在外周血单个核细胞与PC-3肿瘤细胞共培养时,照射组在培养至96h时,γ射线照射外周血单个核细胞对前列腺癌细胞的杀伤作用明显增强,杀伤活性明显增加。结论:γ射线辐照对某些外周血单个核细胞有损伤作用,但是γ射线辐照的外周血单个核细胞其杀伤肿瘤细胞活性增强。Objective: To observe the killing effects of peripheral blood mononuclear cells (PBMCs) irradiated by γ ray on cultured human prostate tumor cell line PC-3. Methods: The experiment cells were divided into PC-3 cell control group, PBMCs control group,PC-3 tumor cells with irradiated or non-irradiated PBMCs co-cultured groups. Radiation dosage was one gray, absorption dose rate was 17Gy/min , acridine orange/ethidium bromide (AO/EB) staining and MTT colorimetric assay were used to observe the killing effects of PBMCs on tumor cells in different period. Results: After culture for 144h , the dead PBMCs in irradiated group were much more than that in non-irradiated PBMCs group. In PBMCs and MKN-28 tumor cells co-cultured group, the killing effects of PBMCs on tumor cells in irradiation group was higher than that in nonradiation group from 96h to 240h. Conclusion:Gamma ray irradiation has damage effect on some PBMCs, but the killing effects of PBMCs irradiated by low dose radiation on tumor cells is increased.
