摘  要:目的:对护心复方药液大孔吸附树脂精制工艺进行评价。方法:以过柱前后复方的HPLC图谱特征峰所代表的化学成分保留率及抗心肌缺血活性相结合的方法对复方药液精制工艺进行评价。结果:护心复方药液过柱后HPLC图谱中特征峰相对保留率达到90%以上,上柱原液与醇洗液HPLC图谱基本一致;大鼠灌胃给予上柱原液、水洗液、醇洗液,上柱原液及醇洗液均可明显改善急性心肌缺血症状,而水洗液药效作用微弱,其活性为:醇洗液>上柱原液>水洗液。结论:该过柱工艺在显著降低得膏率的同时,保留了复方药液HPLC图谱特征峰所代表的化学成分及其抗心肌缺血活性,"谱效结合"的评价方法为中药复方精制工艺提供了参考。Objective:To evaluate the refining process of Huxin compound liquor with macroporous adsorptive resins. Methods:The refining process of the Huxin compound liquor was evaluated by means of the retention rates of main compounds corresponding to main peaks in HPLC fingerprint. The pharmacodynamics was evaluated with the anti-myocardial ischemie activity in vivo. Results:The retention rates of main components were more than 90%. The HPLC chromatograms of the crude liquor and the ethanol extract were similar. Rats were given with crude liquor, water extract and ethanol extract by gavage,respectively. Crude liquor and ethanol extract significantly reduced myocardial ischemia induced by injury, and their activities were in sequence of ethanol extract,crude liquor and water extract. Conclusion:This process not only significantly decreased the yield of extract, but also remained the ingredients corresponding to main peaks in HPLC fingerprints and anti-myocardial isehemic activities. Pharmacodynamics combined with fingerprint could provide a reference for evaluating the refining process of herbal compound.
