摘 要:目的:提高抗肿瘤抗生素在发酵液中的含量,并进行分离纯化。方法:采用微生物法、DNA断裂法分析抗肿瘤抗生素在发酵液中的含量,以响应曲面法优化发酵培养基。结果:通过改变发酵培养基的组份及优化其配比,新型产生菌发酵液中抗肿瘤抗生素的含量提高约30倍。在其后的实验中运用优化的培养基发酵,从中分离纯化出高效抗肿瘤抗生素,经初步测定相对分子质量为1367。结论:培养基组份及其配比影响其代谢产物的组成及含量,响应曲面法可以用相对较少的实验获得优化的发酵条件,提高抗肿瘤抗生素的含量。Objective:To optimize the fermentation for improving content of anti-tumor antibiotic in the fermentation broth. Methods:Bioassay and DNA cleavage assay were used to detect content of anti-tumor antibiotic, and response surface methodology to optimize fermentation medium. Results:In the fermentation broth, content of anti-tumor antibiotic was increased by 30-fold through changing the composition of culture medium and optimizing their ratio. In subsequent experiments, we isolated and purified potent anti-tumor antibiotic from optimal fermentation broth. ESI-MS analysis showed that its molecular weight is 1367Da. Conclusion: Composition and the ratio of the culture medium affect the metabolite components and contents. Response surface methodology can be used to optimize fermentation condition in small number experiments to improve content of anti-tumor antibiotic.
