摘 要:目的通过体外试验研究小檗碱与厚朴酚、和厚朴酚联合对THP-1巨噬细胞源性泡沫细胞形成的影响及其干预途径。方法以氧化型低密度脂蛋白(oxLDL)诱导THP-1巨噬细胞泡沫化为模型,采用HPLC-MS测定泡沫细胞中总胆固醇的含量;ELISA法测定巨噬细胞培养上清液中肿瘤坏死因子-α(TNF-α)的含量;FCM检测巨噬细胞CD36的表达。结果经小檗碱与厚朴酚、和厚朴酚联合作用后,泡沫细胞中总胆固醇的含量显著减少,细胞培养上清液中TNF-α分泌量降低,细胞表面CD36表达被抑制。结论小檗碱联合厚朴酚及和厚朴酚抑制巨噬细胞源性泡沫细胞的形成与抗炎和下调脂质的受体CD36表达有关。Objective To study the effects and mechanism of berberine in combination with magnolol and honokiol on the formation of THP - 1 maerophage - derived foam cells. Methods THP - 1 macrophages were induced into foam cells by ox - LDL, and intracellular cholesterol in foam cells was determined by HPLC - MS ; the concentration of tumor necrosis factor -α in the supernate of THP - 1 macrophages was tested by ELISA and cell - surface CD36 level was assessed by flow cytometry. Results Combined treatment of berberine, magnolol and honokiol significantly reduced cholesterol content in foam cells, also inhibited expression of tumor necrosis factor - α and CD36 in THP - 1 macrophage. Conclusion Co - administration of berberine, magnolol and honokiol can inhibit the formation of foam cells probably through, down - regulating the level of tumor necrosis factor - α and CD36.
