摘 要:目的:通过合成水溶性"纳米银/羧甲基壳聚糖"生物敷料,以标准菌株测试其抑(抗)菌性能。方法:通过紫外吸收光谱表征及扫描电子显微镜形貌观察纳米银,以红外吸收光谱表征由氯乙酸法将壳聚糖改性而成的羧甲基壳聚糖,再将羧甲基壳聚糖与纳米银结合,合成水溶性的"纳米银/羧甲基壳聚糖"生物敷料,再用标准菌株测试其抑抗菌性能。结果:合成的纳米银粒径大小为60~100nm,改性后的羧甲基壳聚糖在IR谱图于1635.56cm-1和1385.22cm-1处出现了羧甲基壳聚糖钠盐的特征吸收峰。该"纳米银/羧甲基壳聚糖"敷料对金黄色葡萄球菌、大肠埃希氏菌、铜绿假单胞菌的MIC分别为:0.25mg/ml(其中含纳米银0.05mmol/L),0.5mg/ml(其中含纳米银0.1mmol/L),0.5mg/ml(其中含纳米银0.1mmol/L);2.5mg/ml"纳米银/羧甲基壳聚糖"(含银0.25mmol/L)在M-H培养基上对金黄色葡萄球菌、大肠埃希氏菌、铜绿假单胞菌的抑菌环分别为17mm,11mm,11mm。结论:合成的"纳米银/羧甲基壳聚糖"生物敷料能结合羧甲基壳聚糖和纳米银的优点,具有强大的广谱抗菌性能。Objective:To synthesize a kind of nano-biological dressing of nano-silver/carboxymethyl chitosan and to investigate its anti(ant)-bacterial activity with standard strain. Methods: Carboxymethyl chitosan(CMC) was changed from chitosan (CTS) by the method of chloroaeetic acid and analyzed its structure by infra-red absorption spectroscopy. The water-soluble nano-silver/CMC was achieved by combining nano-silver with CMC. The antibacterial activity of this novel dressing to standard strain was also studied. Results: The infra-red spectrogram of CMC showed the CTS had been modified by chloroacetic acid. The MIC of the nano-silver/CMC to S.aureus,P.eruginusa and E. coil are 0.25mg/ml(the concentration of nano-silver at 0.05mmol/L),0.5mg/ml(the concentration of nanosilver at 0. 1 mmol/L),0.Smg/ml (the concentration of nano-silver at 0. l mmol/L)respectively. The inhibition zone diameters are 17 mm, 1 lmm and 1 lmm in the M-H medium with nano-silver/CMC. Conclusion: Combining nano-silver with CMC, the water-soluble nanosilver/CMC dressing was achieved. This kind of biodressing possessed powerful broadspectrum antibacterial property.
