摘  要:目的建立毛细管电泳法测定灵芝中核苷类成分含量的方法。方法采用Agilent毛细管电泳仪,熔融石英毛细管56cm×75μm,有效长度50cm,自动压力(30mPa)进样2s,检测波长254nm,分离电压27kV,运行缓冲液为0.05mol/L,硼砂-甲醇(82:18)。结果灵芝中主要成分腺苷、鸟苷、尿苷、肌苷分别在2.33~116.6mg/L,2.08—104.2mg/L,2.06~103.0mg/L,1.08~43.2mg/L范围内有良好的线性关系,相关系数r分别为0.9998、0.9997、0.9996、0.9998。加样回收率分别为97.6%、101.2%、102.7%、101.4%。RSD分别为2.1%、1.6%、2.8%、2.3%。结论此法简便、快速,适用于灵芝中核苷类成分含量的同时测定。Objective To establish CE method for making a assay of nucleosines in Ganoderma. Methods Agilent apparatus equipped with a UV detector and a untreated fused- quartz capillary( 56cm × 75μm,50cm effective length)were used. Pressure injection was 30mpa for 2s, and the wavelength of detector was 254nm. 27kV was applied for separation. The running buffer was 0.05mol/L^-1 Sodium borate - Methanol (82:18). Results The Linear ranges of adenosion,guanosion,uridine,inosion lay in the concentration range of 2.33 - 116.6mg/L,2.08 104.2mg/L,2.06 - 103.0mg/L, 1.08 - 43.2mg/L, respectively, r = 0. 9998,0. 9997,0. 9996,0. 9998. The average recoveries were 97.6% ,101.2%, 102.7% , 101.4%, RSD were 2.1% , 1.6% ,2.8% ,2.3% , respectively. Conclusion The method is simple, rapid. It is suitable for the assay of nucleosions in Ganoderma.
