摘  要:目的应用大孔吸附树脂对山茱萸中环烯醚萜苷类成分进行分离纯化,考察大孔树脂纯化山茱萸总苷的工艺参数。方法考察上样量、洗脱剂浓度、洗脱体积、上样浓度等因素对HPD-100大孔吸附树脂纯化山茱萸环烯醚萜苷的影响。以洗脱液中莫诺苷和马钱苷含量作为考察指标,用HPLC法测定其含量,确定最佳工艺参数。结果洗脱溶剂应为50%乙醇,大孔树脂每使用两次需要再生1次,最佳上样浓度为0.25g/ml,洗脱溶剂的用量为8倍的吸附原液体积,吸附容量为10ml。结论采用大孔树脂分离纯化的山茱萸总苷可用于制备环烯醚萜苷制剂。Objective To study the technological parameters of purification of total glycoside from Comus officinalis with macroporous resin. Methods The sample volume, elution concentration, volume and sample concentration were observed and compared with those elution solvents. The purification of iridoid glycosides was evaluated with contents of morroniside and loganin by HPLC methods. Results Eluting solvent was 50% ethanol, the macroporous resin used twice necded to be renewed, the best sample concentration was 0.25 g/ml, the volume of eluting solvent was eight times the of volume of sample solution, the absorption capacity was 10ml. Conclusion Iridiod glycoside can be purified by macroporous. It is a feasible method to produce preparation.
