摘  要:目的观察不同浓度葡萄糖对体外培养的大鼠肾小球系膜细胞表达和合成单核细胞趋化蛋白-1(MCP-1)的影响,探讨MCP-1在糖尿病肾病发生发展中的意义。方法根据含糖浓度将大鼠肾小球系膜细胞分为3组:①A组(5.6mmol/L);②B组(17.5mmol/L)组;③C组(30mmol/L)。RT-PCR检测系膜细胞MCP-1mRNA表达,ELISA测定细胞上清MCP-1浓度。结果①系膜细胞MCP-1mRNA表达:A组呈弱表达;B组24h后表达增强,48h达高峰,与A组比较P<0.01;C组12h时表达已升高,24h持续高水平,与A、B组同时间点比较P<0.01,但48h时表达降低。②上清液MCP-1蛋白浓度:A组MCP-1浓度随时间变化升高不明显;B组MCP-1浓度随时间变化明显升高,与A组比较P<0.01(除12h段外);C组随时间变化进一步升高,48h时仍持续高水平,与A、B组比较P<0.01。结论葡萄糖刺激系膜细胞MCP-1表达和合成在一定范围内增加,并具有时间和浓度依赖性。增高的表达和合成的MCP-1水平可能参与了糖尿病肾病发生发展。Objective To investigate the significance of monocyte chemoattractant protein-1 (MCP-1) in diabetic nephropathy by comparing the levels of MCP-1 which expressed and synthesized in rats mesangial cells (RMCs) with the different concentration of glucose in vitro. Methods RMCs, used as target cells, were divided into control group (5.6 mmol/L,group A),intermediate glucose group (17.5 mmoL/L,group B) and high glucose group (30 mmol/L,group C). The concentration of MCP-1 in supernatants was determined by ELISA and the expression of MCP-1 mRNA in RMCs was detected by RT-PCR. Results The expressions of MCP-1 mRNA were poor in group A, which in group B and C were obviously higher than those in group A (all P 〈0. 01 ) ,and in group C were further than those in group B at the same time( all P 〈 0. 01 ), except for 48 h;The expressions of MCP-1 protein in the supernatant persistently increased in group B and C by comparing with group A ( all P 〈 0. 01 ), except for 12 h at group B. and which in group C were obviously higher than those in group B ( all P 〈 0. 01 ). Conclusion The expression of MCP-1 mRNA and protein synthesis in RMCs can be induced by the glucose in dose-and time-dependent manners and in the certain range. Elevated expressions of MCP-1 mRNA and protein synthesis may be taken an important role in the progression of diabetic nephropathy.
