必需磷脂防治大鼠肝纤维化的实验研究

摘　　要：目的：复制大鼠酒精及ccl4致大鼠肝纤维化模型，观察必需磷脂对其的防治作用。方法：健康雄性wist—ar大鼠60只，随机分为6组，实验持续4周。A组（空白对照组）：生理盐水灌胃，每天1ml／100g体重。B组（模型组）：白酒灌胃，每天1ml／100g体重，同时cct4腹腔内每次注射0．025mg／100g体重，每周2次。C、D、E组（必需磷脂小、中、大剂量组）：在B组基础上预先灌胃必需磷脂相应剂量。F组（阳性对照组）：在B组基础上凯西莱预先腹腔注射。第4周末取血检测ALT、AST、SOD、MDA、GSH—Px等项目。实验结束处死动物，取肝组织行病理检验。结果：酒精及四氯化碳可引起大鼠肝功能损害、脂质过氧化加重，组织学出现肝纤维化表现。给予必需磷脂可明显减轻这些改变。结论：必需磷脂对大鼠酒精及四氯化碳致肝纤维化有明显防治作用。Objective： To reproduce the rat model of liver fibrosis induced by alcohol and tetrachloride carbon, and observe the protective and therapeutic effects. Methods： Sixty male Wistar rats were divided randomly into six groups, and the experiment was continued for 4 weeks. Group A （control group） was perfused gastrically by saline at the dose of 1ml/100g/d, the other groups were perfused gastrically by 56% alcohol at the dose of 1ml/100 g/d, and at the same time, cc14 was injected intraperitoneally at the dose of 0. 025 mg/100g, b. i. W. Group C, D, E （small, middle and large dose of Essentiale） was beforehand perfused gastrically Essentiale. Group F （control group of positive drug） was beforehand injected intraperitoneally by tiopronin. At the end of the 4th week, ALT, AST, ALP, GSH. Px and MDA were examined. When the experiment was over, rats were euthanized and the histopathologic alterations of liver were observed. Results： The 4th week, group B were obviously different from group A, C, D, E, F. Group B showed significant hepfic injury, including increased ALT, AST, MDA and decreased GSH- Px, SOD; cellular necrosis; inflammatory infiltration and liver fibrosis. When compared with group B, the hepatic injury in group C, D, E, were mild. Conclusion： Essentiale has remarkable prevention and cure function to rat liver fibrosis.
