摘 要:应用RT-PCR方法,对从一只野生白眉鸭(Anasquer quedula)中分离出的禽流感病毒分离株A/Gr-ganey/SanJiang/160/2006(H5N2)的HA基因进行了序列的克隆和测定。测序结果显示,克隆的HA基因全长为1731bp,共编码565个氨基酸。核苷酸和氨基酸的最大同源性比较结果表明,该毒株的HA基因与其它31株H5N2亚型禽流感病毒HA基因的同源性为71.0%~98.2%,氨基酸序列同源性为84.0%~98.2%;遗传进化关系表明,该毒株属于欧亚群系。通过血凝素裂解位点的氨基酸序列分析可知,该毒株的HA切割位点上未见到典型的高致病力毒株H5、H7所具有的一系列碱性氨基酸,符合低致病力禽流感毒株的分子特征。The hemagglutinin (HA) gene of A/Garganey/SanJiang,/160/2006(HSN2) isolated from a wild garganey was amplified by RT-PCR and sequenced. The sequencing result showed that the HA complete nucleotide sequence was 1 731 bp, encoding 565 amino acids. Comparative analysis of isolate A/Garganey/SanJiang/160/2006 with some other isolates of HSN2 subtype indicated that the nucleotide homology ranged from 71.0 percent to 98.2 percent and the amino acid homology was between 84.0 percent and 98.2 percent. Analysis of phylogenetic relationships indicates that isolate A/Garganey/ SanJiang/160/2006 belongs to Eurasian lineage. There are not serial basic amino acids at the cleavage site of HA, which is consistent with the characteristics of low pathogenic AIV.
